Utilizing Network Pharmacology to Explore the Possible Mechanism of Coptidis Rhizoma in Kawasaki Disease

Background: The purpose of the research is to identify main active ingredients in Coptidis Rhizoma (CR) and explore possible molecular mechanisms treatment Kawasaki disease (KD). Materials Methods: A total 58 children with KD were randomly divided into a control group Berberine group. therapeutic indicators two groups before after compared. Then, compounds drug targets CR from TCMSP, SWISS, SEA, STITCH collected, targeted genes retrieved DisGeNET, DrugBank, GeneCards databases. network pharmacology approach involved construction, target prediction, module analysis. GO KEGG enrichment analysis performed investigate pathways related for treatments. Finally, protein expression was determined verify core using Western blotting cell experiment. Results: In total, nine compounds, 369 relative targets, 624 collected above database. revealed that 41 might be on KD. biological processes, namely, response hormone, inorganic substance, enzyme-linked receptor signaling pathway, Pathways cancer, Toll-like Pancreatic cancer are most significant. Protein CASP3, PTGS2, SRC upregulated AKT1 ERK downregulated. Conclusion: We provided useful resources understand mechanism potential novel therapy
